    Achievement incentives determine the effects of achievement-motive incongruence on flow experience

    Previous research has shown that incongruence between implicit and explicit achievement motives impairs flow experience. We examined this relationship in a more differentiated manner by arguing that achievement-motive incongruence only exerts negative effects when individuals act in situations in which achievement incentives are present and arouse conflict between the two motives. In non-achievement situations, no negative effects of achievement-motive incongruence on flow experience are expected. Study 1 and Study 2 showed that participants with incongruent implicit and explicit achievement motives reported less flow in achievement- as compared to non-achievement-oriented sport situations. In Study 3, we experimentally manipulated achievement and non-achievement situations. Again, motive incongruence impaired the experience of flow in achievement but not in non-achievement situation

    Binge eating as a consequence of unfulfilled basic needs: The moderating role of implicit achievement motivation

    The present study hypothesized that unfulfilled basic needs for autonomy, competence and relatedness (SDT, Deci and Ryan, Psychol Inq 11:227-268, 2000) are associated with the impulse to eat and with binge eating. In addition, we assumed that individuals with a high achievement motive, who are characterized by high self-control competences, have the same impulse to eat when confronted with unfulfilled basic needs, but are better able to control the impulse to binge eat than individuals with a low achievement motive. In accordance with these hypotheses, unfulfilled basic needs significantly positively predicted the impulse to eat as well as binge eating behavior. As also expected, the achievement motive did not moderate the effect of unfulfilled needs on the impulse to eat, but did influence the effect of unfulfilled needs on binge eating. The results are discussed in terms of a broader debate about the interaction between basic needs and implicit motive

    Context-based Normalization of Histological Stains using Deep Convolutional Features

    While human observers are able to cope with variations in color and appearance of histological stains, digital pathology algorithms commonly require a well-normalized setting to achieve peak performance, especially when a limited amount of labeled data is available. This work provides a fully automated, end-to-end learning-based setup for normalizing histological stains, which considers the texture context of the tissue. We introduce Feature Aware Normalization, which extends the framework of batch normalization in combination with gating elements from Long Short-Term Memory units for normalization among different spatial regions of interest. By incorporating a pretrained deep neural network as a feature extractor steering a pixelwise processing pipeline, we achieve excellent normalization results and ensure a consistent representation of color and texture.     On Your Mark, Get Set, Self-Control, Go: A Differentiated View on the Cortical Hemodynamics of Self-Control during Sprint Start

    Most sports are self-control demanding. For example, during a sprint start, athletes have to respond as fast as possible to the start signal (action initiation) while suppressing the urge to start too early (action inhibition). Here, we examined the cortical hemodynamic response to these demands by measuring activity in the two lateral prefrontal cortices (lPFC), a central area for self-control processes. We analyzed activity within subregions of the lPFC, while subjects performed a sprint start, and we assessed if activation varied as a function of hemisphere and gender. In a counterbalanced within-subject design, 39 participants (age: mean (M) = 22.44, standard deviation (SD) = 5.28, 22 women) completed four sprint start conditions (blocks). In each block, participants focused on inhibition (avoid false start), initiation (start fast), no start (do not start) and a combined condition (start fast; avoid false start). We show that oxyhemoglobin in the lPFC increased after the set signal and this increase did not differ between experimental conditions. Increased activation was primarily observed in ventral areas of the lPFC, but only in males, and this increase did not vary between hemispheres. This study provides further support for the involvement of the ventral lPFC during a sprint start, while highlighting gender differences in the processing of sprint start-induced self-control demands

    A high implicit affiliation motive does not always make you happy: A corresponding explicit motive and corresponding behavior are further needed

    Satisfaction of the implicit affiliation motive is known to be positively related to emotional well-being, whereas the frustration of the implicit affiliation motive leads to impairment of well-being. In the present research we specified two conditions that are responsible for the satisfaction and frustration of the implicit motive. Referring to research on the congruence of implicit and explicit motives, we assumed that a corresponding explicit affiliation motive leads to satisfaction of the implicit motive. Corresponding affiliation behavior constitutes the second condition. Three studies confirmed the hypothesis that both conditions must be fulfilled in order to positively connect the implicit affiliation motive to emotional well-being. Participants with high implicit and explicit affiliation motives and who additionally showed a large amount of affiliation behavior reported the lowest negative affectivity and the highest life satisfaction compared to participants who lacked one of the condition

    Boredom is the root of all evil-or is it? A psychometric network approach to individual differences in behavioural responses to boredom.

    Functional accounts of boredom propose that boredom serves as an impartial signal to change something about the current situation, which should give rise to adaptive and maladaptive behaviour alike. This seemingly contrasts with research on boredom proneness, which has overwhelmingly shown associations with maladaptive behaviour. To shed light on this discrepancy, we disentangled boredom proneness from individual differences in (i) the urge to avoid and escape boredom and (ii) adaptive and maladaptive ways of dealing with boredom by developing corresponding trait scales. In a study with N = 636 participants, psychometric network modelling revealed tight associations between boredom proneness and less adaptive and (especially) more maladaptive ways of dealing with boredom. However, its associations with the urge to avoid and escape boredom were rather weak. Importantly, a higher urge to avoid and escape boredom was linked not only to more maladaptive but also to more adaptive ways of dealing with boredom. This pattern of results was robust across various specific behaviours that have previously been linked to boredom. Our findings provide novel evidence for functional accounts of boredom from an individual difference perspective, cautioning against a shallow view of boredom as being associated with purely maladaptive behaviour

    An Intracellular Nanotrap Redirects Proteins and Organelles in Live Bacteria

    Owing to their small size and enhanced stability, nanobodies derived from camelids have previously been used for the construction of intracellular "nanotraps," which enable redirection and manipulation of green fluorescent protein (GFP)-tagged targets within living plant and animal cells. By taking advantage of intracellular compartmentalization in the magnetic bacterium Magnetospirillum gryphiswaldense, we demonstrate that proteins and even entire organelles can be retargeted also within prokaryotic cells by versatile nanotrap technology. Expression of multivalent GFP-binding nanobodies on magnetosomes ectopically recruited the chemotaxis protein CheW(1)-GFP from polar chemoreceptor clusters to the midcell, resulting in a gradual knockdown of aerotaxis. Conversely, entire magnetosome chains could be redirected from the midcell and tethered to one of the cell poles. Similar approaches could potentially be used for building synthetic cellular structures and targeted protein knockdowns in other bacteria. IMPORTANCE Intrabodies are commonly used in eukaryotic systems for intracellular analysis and manipulation of proteins within distinct subcellular compartments. In particular, so-called nanobodies have great potential for synthetic biology approaches because they can be expressed easily in heterologous hosts and actively interact with intracellular targets, for instance, by the construction of intracellular "nanotraps" in living animal and plant cells. Although prokaryotic cells also exhibit a considerable degree of intracellular organization, there are few tools available equivalent to the well-established methods used in eukaryotes. Here, we demonstrate the ectopic retargeting and depletion of polar membrane proteins and entire organelles to distinct compartments in a magnetotactic bacterium, resulting in a gradual knockdown of magneto-aerotaxis. This intracellular nanotrap approach has the potential to be applied in other bacteria for building synthetic cellular structures, manipulating protein function, and creating gradual targeted knockdowns. Our findings provide a proof of principle for the universal use of fluorescently tagged proteins as targets for nanotraps to fulfill these tasks
